Daptomycin is a cyclic lipopeptide antibiotic that has a unique mechanism of action. It binds to the bacterial cell membranes, which causes rapid depolarization of the membrane due to K efflux and associated disruption of DNA, RNA, and protein synthesis resulting in rapid concentration-dependent bacterial death. Daptomycin has broad-spectrum activity against gram-positive bacteria. Cross-resistance with other classes of antimicrobials does not occur thus, daptomycin retains activity against multidrug-resistant pathogens, such as vancomycin - and methicillin-resistant Staphylococcus aureus, vancomycin -resistant enterococci, and pneumococci with reduced penicillin sensitivity. |
